摘要:通过动态提取-超滤-纳滤操作分离金毛狗脊中的总黄酮。以去离子水为溶剂,在十二烷基硫酸钠(SDS)的辅助下动态循环提取金毛狗脊黄酮,并通过超滤去除大分子杂质,再用纳滤对料液进行浓缩。优化的工艺条件为:超滤操作压力0.14 MPa,温度45℃,溶液p H 6,操作时间90 min,此时超滤回收率61.30%,截留率18.52%;纳滤操作压力0.4 MPa,温度45℃,溶液p H 6,操作时间90 min,此时料液浓度提高2.190倍,纳滤回收率68.29%,截留率53.92%。与大孔树脂纯化实验相比,文中采用的分离方法能节省操作时间、提高操作效率,同时能有效提高总黄酮的浓度。
The dynamic extraction-uhrafiltration-nanofiltration operation was used to purify the flavonoids in Cibotium barometz. The flavonoids were extracted by the dynamic circulating water solution with the assistance of sodium dodecyl sulfate (SDS), then the macromolecular impurities were removed by uhrafiltration and the feed was concentrated by nanofihration membrane finally.The ultrafihration yield of 61.30% and the retention rate of 18.52% were obtained under the following optimal conditions: ultrafihration pressure of 0. 14 MPa, temperature of 45 ℃, solution pH of 6, and operation time of 90 min. The nanofiltration yield reached 68.29% and the retention rate of 53.92% under nanofihration pressure of 0. 4 MPa, temperature of 45 ℃ , solution pH of 6, and operation time of 90 min with the trapped fluid flavonoid concentration raised to 2. 190 times than the original one. Compared with the nanofiltration and maeroporous resin purification, results showed that the succession operation method was time-saving and efficient, and the flavonoids was effectively concentrated.
