摘要:采用对倍稀释法测定壳聚糖对金黄色葡萄球菌、大肠杆菌、绿脓杆菌和白色念珠菌的最小抑菌浓度(MIC)和最小杀菌浓度(MBC),并用定量法测定壳聚糖与溶菌酶复合体系的杀菌率,与单一成分的壳聚糖或溶菌酶进行对比,最后考察了壳聚糖和溶菌酶复合体系的稳定性,并对其抑菌机理进行初步研究。结果表明:2g/L壳聚糖虽然对金黄色葡萄球菌、大肠杆菌和铜绿假单胞菌有一定的杀菌率,但对白色念珠菌的杀菌率还不到50%,而溶菌酶质量浓度在5~20g/L对受试菌无明显抑菌作用。将2g/L壳聚糖和10g/L溶菌酶复配后,对受试菌的杀菌率优于单独使用壳聚糖或单独使用溶菌酶,尤其是对白色念珠菌的杀菌力明显提高;将复配液样品置于54℃恒温箱内14d,杀菌率略有降低,但仍然保持在90%以上,说明复配体系稳定性良好。
Minimal inhibitory eoncentration(MIC) and minimal bactericidal coneentration(MBC) of chitosan to Staphylococcus aureus ( St. aureus) , Escherichia coli (E. coli) , Pseudomonas aeruginosa ( P. aeruginosa ) and Candida albicans (M. albicans) were determined by using the tube dilution method. The bactericidal rate of chitosan and lysozyme composite system was tested by using quantitative method, comparing with that of chitosan or lysozyme. Finally, the stability of the composite system was examined and the mechanism was discussed preliminarily. The results showed that 2 g/L chitosan had a certain bactericidal rate to St. aureus, E. coli and P. aeruginosa, while a rather low rate to M. albicans which was lower than 50;. Lysozyme didn't have obvious antibacterial effect to the tested bacteria in the concentration range of 5--20 g/L. After compositing 2 g/L chitosan and 10 g/L lysozyme , there had an obvious bacteric y better bactericidal rate than before, especially to M. albicans. The only slight drop of dal rate for the composite system after 14 d in a 54 ℃ condition indicated favorable stability.
