摘要:为全面、客观评价不同超声提取条件下灵芝菌丝体水提液(GLMw)的体外抗氧化活性,利用超声辅助提取法对灵芝菌丝体的水溶性抗氧化成分进行提取工艺研究,分别考察液固比、超声功率、超声时间、超声温度的影响,在此基础上进行4因素5水平星点设计效应面优化。通过熵权法对30组灵芝菌丝体水提液的1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除率、2,2′-联氮双(3-乙基苯并噻唑啉-6-磺酸)二铵盐(ABTS)自由基清除率、羟自由基清除率进行赋权,分别得到3种自由基清除率的权重,以加权后计算得到的综合抗氧化活性为效应值进行多元回归拟合,得到最佳提取工艺条件。结果表明:在液固比50 mL/g、超声温度40℃、超声功率156 W、超声时间60 min条件下能够得到综合抗氧化活性最高的灵芝菌丝体水提液,该条件下的预测值为63.48%,与实验值62.06%接近(相对标准偏差RSD<3%)。
Ultrasonic assisted extraction method was used to extract water soluble antioxidant components from Ganoderma lucidum mycelium for evaluating the antioxidant activity of water extracts from Ganoderma lucidum mycelium under different ultrasonic extraction conditions in vitro comprehensively and objectively.The effects of liquid-solid ratio,ultrasonic power,time and temperature were investigated,and then the four factors-five levels central composite design response surface methodology was used to optimize the extract process.The 1,1-diphenyl-2-picrylhydrazyl(DPPH),2,2′-bis(3-ethylbenzothiazoline-6-sulfonic acid)diamimium salt(ABTS)and hydroxyl radical scavenging rate of 30 groups of Ganoderma lucidum mycelium aqueous extracts were weighed by entropy method,and the objective weights of the three methods were obtained respectively.The optimal extraction conditions were obtained by multiple regression fitting using the comprehensive antioxidant activity calculated after weighting as the effect value.Results showed that under the conditions of liquid-solid ratio of 50 mL/g,ultrasonic power of 156 W,ultrasonic time of 60 min and ultrasonic temperature of 40℃,the water extract of Ganoderma lucidum mycelium with the highest comprehensive antioxidant activity could be obtained.Under these conditions,the predict comprehensive antioxidant value was 63.48%,which was close to the experimental value of 62.06%(relative standard deviation RSD<3%).
