摘要:本文从成年小鼠脑基因库中克隆了两种独特的胶源神经营养因子(glialcelline-derivedneurotrophicgrowthfactor,GDNF)转录产物(GDNF-α和GDNF-β)。GDNF-β除了在开放阅读框架区(ORF)有78bp的缺失外,其余与GDNF-α相同。由小鼠GDNF-α基因推导得的氨基酸顺序与大鼠和人的氨基酸顺序具有高度同源性。对小鼠不同组织的反转录PCR(RT-PCR)实验表明在这些组织中广泛存在着这两种形式的GDNF。GDNF-α至少由两个外显子编码组成。
We have cloned two unique forms of glial cell linederived neurotrophic growth factor transcripts (GDNFα and GDNFβ) from adult mouse brain. The GDNFβ form is similar to GDNFα except for a 78 bp deletion in the ORF. The deduced amino acid sequences of the mouse GDNFα are highly homologous to those of rat and human GDNF. Reversetranscription PCR from various mouse tissues showed extensive expression of both forms of GDNF. GDNFα is coded by at least two distinct exons. The 3′UTR is entirely encoded in a single exon.
