摘要:对桑叶水提取的粗多糖在离子交换纤维素上的吸附量进行了考察,然后利用离子交换纤维素柱色谱将粗多糖分为4个电荷性质不同的亚组分SD0、SD0.1、SD0.2、SD0.3。通过桑叶粗多糖在离子交换纤维素的吸附解吸行为可知,桑叶多糖主要以酸性多糖的形式存在。其中主要组分SD0.2进一步采用Sephacryl S-200凝胶介质进行纯化,采用苯酚-硫酸法和高效凝胶过滤色谱法同时进行检测,得到均一多糖SDS-1和SDT-1。SDS-1的Mw=8.9×104,Mn=7.5×104,分散度D=1.18;SDT-1的Mw=1.54×104,Mn=1.42×104,D=1.08。
The process of separation for crude polysaccharides from Mulberry Leaves by the ionic exchange cellulose chromatography was studied. Results showed that the adsorption capability of DEAE- cellulose for polysaccharide was 20 mg/mL and the crude polysaccharide could be separated into four parts SD0, SD0.1, SD0.2, SD0.3 with different electrical charge which indicated the polysaccharide from mulberry leaves mainly existed in acidic polysaccharide. High performance liquid gel filtration chromatography indicated the four parts were all inhomogeneous. Furthermore, the major component SD0.2 was separated into two kinds of homogeneous pure polysaccharide SDS-1, SDT-1 by Sephacryl S-200 column chromatography. The weight molecular weight, number weight molecular weight, polydispersity for SDS-1 and SDT-1 were 8. 9 × 10^4 , 7.5 × 10^4 , 1.18 and 1.54 × 10^4 , 1.42 × 10^4 , 1.08 respectively.
