摘要:目的:研究高速逆流色谱分离制备沉香中2-(2-苯乙基)色酮类活性成分的方法。方法:采用氯仿-甲醇-水(4∶2.6∶2.4)为两相溶剂体系,上相为固定相,流速1.2 mL/min,正向转速900 rpm。结果:利用高速逆流色谱法从沉香95%乙醇粗提物中一次性分离得到两个2-(2-苯乙基)色酮衍生物——沉香四醇(agarotetrol,AH1)和4′-甲氧基沉香四醇。所得两分离峰经EI-MS、1H-NMR和13C-NMR鉴定确认。HPLC检测纯度可达99.0%以上。结论:高速逆流色谱法对于沉香中结构相近的2-(2-苯乙基)色酮衍生物具有理想的分离效果,产物纯度高。
AIM:To establish the method of separating 2-(2-Phenylethyl)chromone derivatives from agalwood by high-speed counter-current chromatography(HSCCC).METHODS:Chloroform-methanol-water(4:2.6:2.4,v/v/v) was used as the two-phase solvent system.The upper phase was used as the stationary phase,the revolution speed was 900 rpm and the flow rate was 1.2 mL/min.RESULTS:A preparative HSCCC method was successfully established for separating agarotetrol and 4′-methoxyagarotetrol from 95% ethanol extract of Aquilaria sinensis(Lour.)Gilg.Identification of two constituents was carried out according to its EI-MS、1H-NMR and 13C-NMR da-ta.The purity of agarotetrol and 4′-methoxyagarotetrol was 99.0%,as determined by HPLC.CONCLUSION:HSCCC is successfully used for the separation and purification of two main 2-(2-Phenylethyl)chromone derivatives which has similar structure from agalwood with ideal purity.
