摘要:从螺旋藻中分离纯化的Fe-SOD平均比活为4576U/mg,总活力回收率50%,纯度达电泳纯。SDS-PAGE测定纯化的Fe-SOD亚基相对分子质量为21ku,每分子亚基含0.7个Fe原子;在Tris-盐酸缓冲液中的最适pH为8.2、最适温度为25°C;在紫外区279nm有最大光吸收,酶的pH稳定范围为6~11,65°C以上迅速失活,该酶对H2O2敏感,在5mmol/LH2O2中迅速失活,而在φ=0.1的乙腈和5mmol/L叠氮化钠中稳定。
FeSOD in Spirulina platensis is separated and purified, the specific activity of the purified FeSOD is 4 576U/mg and it is identified as electrophoresis purity with SDSPAGE. Its subunit molecular mass is 21ku and with 0.7 Fe atom per subunit. The enzyme exhibits one absorption peak at 279 nm. The optimum pH in TrisHCl buffer is 8.2 and the optimum temperature is 25°C. The enzyme remains stable in the range of pH=6~11 and below 65°C. Its activity could be inhibited rapidly by 5 mmol/L H2O2 but unaffected by φ=0.1 CH3CN and 5mmol/L NaN3.
