摘要:曾利用同源重组技术构建了稳定的必特螺旋霉素基因工程菌。本研究在30L全自动发酵罐从菌体生长,糖、氮源利用及发酵效价增长情况考察了必特螺旋霉素基因工程菌的发酵过程,研究了发酵过程摄氧率(OUR)、二氧化碳释放率(CER)、溶解氧(DO)等在线参数的变化,着重研究了不同溶氧水平对必特螺旋霉素发酵的影响。结果表明溶氧水平尤其是发酵前期溶氧水平对必特螺旋霉素发酵影响至关重要,前期临界氧浓度应控制在25%左右。本研究为必特螺旋霉素发酵放大工艺研究提供了依据。
Biotechmycin is a new antibiotic produced by a stable genetically-engineered strain WSJ-1 which is constructed through gene transformation and homologous recombination. Biotechmycin fermentation course was investigated by monitoring the sugar and nitrogen consumption as well as the mycelium growth. The effect of dissolved oxygen (DO), especially the level of DO at the beginning of the fermentation, on biotechmycin production was studied. Based on determination of on-line and off-line parameters, the variations of on-line parameters, such as oxygen uptake rate (OUR), carbon dioxide exchange rate (CER), DO etc. were investigated by the changes of agitation speed in different degree during the fermentation course. The critical DO level in the fermentation scale-up process of biotechmycin was proposed.
