摘要:目的测定从板蓝根粗多糖中分离纯化得到的板蓝根糖肽的氨基酸和单糖组成成分。方法 70%醇沉粗多糖经柱层析分离得板蓝根糖肽,其单糖组成采用1-苯基-3-甲基-5-吡唑啉酮(PMP)衍生化高效液相色谱法测定。采用响应面试验优化板蓝根糖肽的肽链酸水解条件,使用柱前在线衍生高效液相色谱法测定其氨基酸组成。结果单糖组成为:甘露糖∶鼠李糖∶半乳糖醛酸∶葡萄糖∶半乳糖∶木糖∶阿拉伯糖=132.6∶5.0∶1.0∶7.6∶7.0∶23.8∶76.8,总质量百分数为94.5%。氨基酸组成测定的最佳水解条件是在104℃下用10 mol/L的盐酸水解17 h。测得板蓝根糖肽的氨基酸组成为:天门冬氨酸∶谷氨酸∶丝氨酸∶组氨酸∶苏氨酸∶精氨酸∶丙氨酸∶缬氨酸∶苯丙氨酸∶异亮氨酸∶亮氨酸∶赖氨酸=3.8∶3.8∶2.5∶0.7∶2.5∶1.1∶2.9∶1.9∶0.6∶1.0∶1.5∶1.3,总质量百分数为5.4%。结论本研究为阐明板蓝根糖肽的一级结构奠定了基础。
AIM To determine the compositions of amino acids and monosaccharides in glycopeptide(ISP)obtained from Isatidis Radix.METHODS ISP was obtained from crude polysaccharide centrifuged from 70%Isatidis Radix ethanolic extract.The composition of monosaccharides was determined by HPLC with pre-column derivatization using 1-phenyl-3-methyl-5-pyrazolone.Response surface analysis method was used to optimize the hydrolysis conditions of the amino acids in ISP.Then,an online pre-column derivatization HPLC method was developed to determine the composition of amino acids.RESULTS The composition of monosaccharidesis consisted of mannose ∶ rhamnose ∶ galacturonic acid ∶ glucose ∶ galactose ∶ xylose ∶ arabinose(132.6 ∶ 5.0 ∶ 1.0 ∶ 7.6 ∶ 7.0 ∶23.8 ∶ 76.8),and the monosaccharides reached 94.5% of ISP in mass.The optimized hydrolysis conditions of amino acids in ISP were dissolved in 10 mol / L aqueous hydrochloric acid for 17 h at 104 ℃.The molar ratio of the hydrolyzed amino acid was of Asp ∶ Glu ∶ Ser ∶ His ∶ Thr ∶ Arg ∶ Ala ∶ Val ∶ Phe ∶ Ile ∶ Leu ∶ Lys(3.8 ∶ 3.8 ∶2.5 ∶ 0.7∶ 2.5 ∶ 1.1 ∶ 2.9 ∶ 1.9 ∶ 0.6 ∶ 1.0 ∶ 1.5 ∶ 1.3),and nearly 5.4% of the total mass.CONCLUSION This study provides a possible support for elucidating the primary structure of glycopeptides.
