摘要:NOVELTY - Human serum alpha -ketone glutaric acid directly fixed quantity detecting reagent box comprises a serum tube, hepes buffer solution, reductive coenzyme, enzyme stabilizers, fluorescent dye and a alpha-KG, where reducing coenzyme contains reduced coenzyme Q10, nicotinamide adenine dinucleotide, or nicotinamide adenine dinucleotide phosphate; the enzyme stabilizer comprises trehalose, mannitol, glycerine, vitamin C, or bovine serum albumin. USE - Used as human serum alpha -ketone glutaric acid directly fixed quantity detecting reagent box. ADVANTAGE - The kit can rapidly and efficiently detect alpha-ketone glutaric acid in liver cirrhosis and liver cancer patients serum, is economical, has simple operation, fast high-flux characteristic, has specific selectivity and high stability, can obviously improve the repeatability on the operation, is suitable as tool for diagnosing liver cirrhosis, liver cancer, evaluation for early detection and treatment effect of liver, and has important meaning and wide application prospect. DETAILED DESCRIPTION - INDEPENDENT CLAIM is also included for: (1) detecting alpha-ketone glutaric acid in serum, comprising taking the kit in the refrigerator of 20 degrees C, at the same time, preparing human serum to be tested, dispensing tube of the serum, hepes buffer solution, reductive coenzyme, enzyme stabilizers, fluorescent dye, alpha-ketone glutaric acid and human serum are added into black semi-quantitative 96 plate, in the enzyme-labeling detecting the fluorescence intensity change, according to the change in fluorescence value of microplate reading, using standard detection method of alpha-ketone glutaric acid, calibrating the concentration of alpha alpha-KG in serum; and (2) use of directly fixed quantity detecting device in preparing human serum alpha-ketone glutaric acid.
