摘要:目的:考察环境及内源条件对克雷伯氏肺炎杆菌(Klebsiella pneumoniae)质粒消除的影响。方法:采用高温、SDS等环境手段以及基因组基因敲除等分子生物学手段消除K.pneumoniae的质粒。结果:采用高温和SDS同时处理的方法,在最适的条件下质粒消除率可达41.7%。在考察荚膜基因缺失对质粒消除的影响时发现,相同的作用条件下荚膜基因的缺失降低了质粒的消除效率,并在此进一步考察了其他基因缺失对质粒消除的影响。结论:高温和SDS复合处理可以显著提高质粒消除的效果。基因组基因的缺失对质粒的消除有影响。
Objective: The impact of various conditions on plasmid elimination in KlebsieUa pneumoniae was systematically researched. Method: The plasmid of K. pneumoniae was eliminated by high temperature and SDS as well as gene knockout of K. pnumoniae genome. Result: By culturing in high temperature incorporated with adding SDS, the efficiency of plasmid elimination reached up to 41.7%. Under the same conditions, it was found that the deletion of capsular gene of genome reduced the efficiency of plas-mid elimination in Klebsiella pneumoniae. In the further research, the effect of the other gene deletion of genome on the plasmid elimination was studied. Conclusion:The efficiency of plasmid elimination could significantly improved by culturing in high temperature incorporated with adding SDS. And it was found an effect of lack of the genome gene on plasmid elimination in K. pneumoniae.
