摘要:建立一种测定丙酮酸发酵液中丙酮酸含量的方法。采用有机酸离子排斥柱,TSKgel OApak-A为分析柱,TSKgel OApak-P为保护柱;柱温:40℃;流动相:4.5mmol/L H2SO4,流速0.8mL/min;检测波长:210nm。丙酮酸在42.45~2122.3mg/L范围内,峰面积与样品浓度有良好的线性关系(A=4.7278c+31.6296,r=0.9999),检出限为4.6mg/L。发酵液中丙酮酸含量在5.1%左右,丙酮酸的回收率为97.11%~99.21%。
An ion exclusion chromatographic method for analysing pyruvate in pyruvic acid fermentation broth was established. The separation was performed on the TSKgel OApak -A ion exclusion analytical column and TSKgel OApak -P guard column with 4.5 mmol/L H2SO4 at a flow of 0.8 mL/min. The column temperature was dO^C and the detection wavelength was at 210 nm. The pyruvic acid linear equation was A = 4. 727 8c + 31. 629 6, and the correlation coefficient was 0.999 9. The detection limit was 4.6mg/ L, and the average recoveries were 97.11% -99.21%.
