摘要:以相同条件下栽培获得的沪农系列7个不同品种的灵芝(Ganoderma lucidum)子实体作为研究材料,分析比较其总多糖、核苷、三萜和糖醇的含量、多糖重均分子量分布特征差异以及刺激RAW 264.7巨噬细胞释放NO的活性。结果表明:不同品种灵芝总多糖含量在1.15%~5.11%之间,含量较高的3个品种由高到低为‘沪农芝7号’(5.11%)、‘沪农芝8号’(2.36%)、‘沪农灵芝4号’(2.07%)。不同灵芝品种粗多糖分子量分布存在差异,‘沪农灵芝4号’主要含有重均分子量为3.38×10^(6)、4.24×10^(6)、4.38×10^(3)g·mol^(-1)的三个多糖组分,‘沪农灵芝1号’主要含有重均分子量为1.01×10^(7)和8.03×10^(3)g·mol^(-1)的两类多糖组分,‘沪农芝7号’主要含有重均分子量为4.50×10^(5)、6.20×10^(3)g·mol^(-1)的两个多糖组分,其余4个品种均含有重均分子量为(4.56×10^(3)~5.47×10^(3))g·mol^(-1)的多糖组分。7个灵芝新品种子实体中均含有胞苷、尿苷、鸟苷,其中‘沪农灵芝4号’的总核苷含量最高(1456.5μg·g^(-1));总三萜的含量为2241.93~5123.54μg·g^(-1),最高的为‘沪农灵芝1号’(5123.54μg·g^(-1));不同灵芝品种的糖醇含量存在一定差异,主要含有阿拉伯糖醇和甘露糖醇,赤藓糖醇含量较低,均低于0.9 mg·g^(-1)。七种灵芝子实体的粗多糖浓度在50~500μg·mL^(-1)范围内均能增强RAW 264.7巨噬细胞NO的释放量,其中‘沪农灵芝5号’的活性最强。研究结果将为沪农系列灵芝新品种的开发利用和推广应用提供参考。
Seven cultivars of Ganoderma lucidum‘Hunong’series were cultivated under the same conditions and the resultant fruiting bodies were determined and compared for total polysaccharide content,nucleoside content,triterpenoid content,sugar alcohol content,molecular weight distribution of polysaccharides,and stimulation activity on RAW 264.7 macrophages to release NO.The results showed that the total polysaccharide content of different‘Hunong’cultivars ranged from 1.15%to 5.11%,and the top three high polysaccharide content cultivars were G.lucidum‘Hunong No.7’(5.11%),G.lucidum‘Hunong No.8’(2.36%)and G.lucidum‘Hunong No.4’(2.07%).There were differences in the molecular weight distribution of crude polysaccharides from different G.lucidum cultivars.G.lucidum‘Hunong No.4’mainly contained three polysaccharide components with weight-average molecular weights of 3.38×10^(6)g·mol^(-1),4.24×10^(6)g·mol^(-1)and 4.38×10^(3)g·mol^(-1),respectively.G.lucidum‘Hunong No.1’mainly contained two polysaccharide components with weight-average molecular weights of 1.01×10^(7)g·mol^(-1)and 8.03×10^(3)g·mol^(-1),respectively.G.lucidum‘Hunong No.7’mainly contained two polysaccharide components with weight-average molecular weights of 4.50×10^(5) g·mol^(-1)and 6.20×10^(3)g·mol^(-1),respectively.The remaining four cultivars contained a polysaccharide component of 4.56×10^(3)g·mol^(-1)-5.47×10^(3)g·mol^(-1).Fruiting bodies of all seven new cultivars were found to contain cytidine,uridine,and guanosine.G.lucidum‘Hunong No.4’showed the highest nucleoside content(1456.5μg·g^(-1)).The triterpenoid content of the seven cultivars ranged from 2241.93μg·g^(-1)to 5123.54μg·g^(-1),and G.lucidum‘Hunong No.1’showed the highest content.For sugar alcohols,the seven cultivars mainly contained arabitol and mannitol.Erythritol was detected at a low level(<0.9 mg·g^(-1)).The water-soluble polysaccharides extracted from the seven new cultivars enhanced NO release from RAW264.7 macrophages at 50-500μg·mL^(-1),and that of G.lucidum‘Hunong No.5’showed the highest activity.The results provided a scientific basis for the development and utilization of the new G.lucidum cultivars of‘Hunong’series.
