摘要:利用自行设计和制造的微流控两相和三相装置实现对聚乳酸-羟基乙酸共聚物[poly(lactic-co-glycolic acid),PLGA]微球的可控制备,并对其性能进行研究,调控微流控操作参数,制备PLGA微球,并以传统乳化/溶剂挥发法作对照同时制备PLGA微球,对所得微球的形貌及粒径进行分析。结果表明,乳化/溶剂挥发法得到的微球分散性差,大小不均一,离散系数(coefficient of variation,CV)为44.13%,而微流控法得到的微球分散性极好,大小均一,且CV仅为2.73%(两相)和1.93%(三相),说明微流控法能够制备单分散性很高的PLGA微球,有着乳化/溶剂挥发法无法比拟的优势。
The controllable preparation of poly( lactic-co-glycolic acid)( PLGA) microspheres is achieved by selfdesigned and self-made two-fluid-channel device and three-fluid-channel microfluidics device.The performance of PLGA microspheres is studied. The microfluidics parameters are adjusting and varying in the preparation process. PLGA microspheres are also prepared by traditional emulsion/solvent evaporation method,and are taken as a reference.Morphology and size distribution of the PLGA microspheres prepared by both methods are investigated. It is shown that PLGA microspheres prepared by emulsion/solvent evaporation method has serious aggregation and a wide size distribution with a coefficient of variation( CV) being 44. 13%,whereas those prepared by two-fluid-channel microfluidics device and three-fluid-channel microfluidics device both have good dispersive performance,narrow size distribution,and CV of 2. 73%( two-fluid-channel device) and 1. 93%( three-fluid-channel device),respectively. It indicates that using microfluidics can manufacture highly mono-dispersive PLGA microspheres without aggregation.Hence,it is possible to achieve the precise control over the preparation of PLGA microspheres.
