摘要:本文利用氯化钙和脱乙酰几丁质改进卡拉胶载体用于固定化G.oxydans和B.cereus活细胞取得理想效果.脱乙酰几丁质与卡拉胶可以形成韧性和通透性良好的网络结构,改善了固定化细胞的各种性能.凝胶中的细胞密度可高达3.25×10~3个细胞/ml凝胶,固定化细胞可反复利用11批,每批发酵收率均可维持在65%左右。
The properties of carrageenan as the carrier for coimmobilizing Gluconobacter oxydans and Bacillus cereus living cells, can be improved by the addition of CaCl2 and Chitosan in producing 2-Keto-L-gulonic acid (2-KLG, Vitamin C precursor). The improvements include the mechanical strength, the capacity of producing 2-KLG, cell stability. It is found that a hard cover with good toughness and permeability forms on gel surface due to the reaction of Chitosan upon carrageenan. Contrast experiments show that cell concentration can be raised up to 3.25*108 cells/ml gel.The coimmobilized living cells can be used for batch fermentation for 11 times with approximate 65% yields and shortened fermentation period.
