摘要:应用数学模型方法描述重组质粒由于拷贝数的增加以及基因表达产物——乙型肝炎表面抗原在受体细胞内积累造成对Leu^+表型细胞生长的阻遏作用。模拟丢失重组质粒的细胞生长规律,给出其形成后可能的年龄-时间分布。根据这两类不同表型细胞同时生长的轨迹,得到重组质粒丢失频率与Leu^-表型细胞生长的关系。因此,可把发酵过程中基因工程酵母丧现出重组质粒的不稳定归于两种作用:重要的是Leu^+表型细胞分裂时具有不对称的质粒分配,其次是在丰富培养基中这两类不同表型细胞具有不同的生长速率。
A mathematical model is suggested to describe effects of intracellular inhibition on growth of the Leu^+ phenotype cells. The inhibition term depends upon the increase of recombinant plasmid concentration and the accumulated amount of hepatitis B surface antigen protein, the expressed gene product. Simulating the growth of plasmid-free cells gives possible age-time distribution corresponding to their formation. According to the growth trajectores of these two different phenotype cells, a relation between Leu^- phenotype cell growth and recombinant plastaid loss frequency is obtained.
