摘要:通过对螺旋链霉菌的中间代谢产物及有机酸的分析 ,建立了螺旋霉素生物合成的动态流量分布图 ,并分析了螺旋霉素生物合成代谢网络的节点。分析认为 ,大环成环步骤和 FO- 1步骤在发酵中前期有一定的限速影响 ;同时由于 FO- 2、NSP- 2有一定的积累 ,若能减少两者的形成 ,将对提高发酵效率有积极的意义。摇瓶实验验证了添加葡萄糖对螺旋霉素生物合成代谢网络有影响 ,发酵效价提高了 2 6 .8%。
By analyzing the organic acids and other intermediary metabolic products during spiramycin biosynthesis, the dynamic flux distribution profiles were established. Based on these findings,the knot points of the metabolic networks of the bioconversion of spiramycin were investigated. Results revealed that the 14 member ring forming process and the transform from FO 1 to NSP 1 were critical during the fermentation prophase. The efficiency of spiramycin production was closely related to the reduction of FO 2 and NSP 2 which were accmulated simultaneously during ring formation. It was conformed in the flask experiments, the addition of glucose produced a positive effect on the biosynthesis of spiramycm,with an increased potency by 26.8%.
