摘要:通过高温蒸汽浸提法提取黑木耳多糖。在模拟体外消化后,黑木耳多糖发生降解。通过刚果红的结果证明消化后黑木耳多糖中的三股螺旋结构消失。FT-IR光谱和糖醛酸检测证明了多糖中的糖醛酸被水解。之后将提取的黑木耳多糖体外消化产物对RAW264.7细胞进行处理,发现经胃肠液消化后的黑木耳多糖是通过刺激TLR4受体介导的JNK以及ERK和NF-κB信号通路,从而刺激巨噬细胞增殖,诱导NO的释放并增加巨噬细胞的吞噬活性。可见黑木耳多糖作为免疫调节剂具有良好的前景。
In this study,the Auricularia auricula polysaccharide was extracted by high-temperature steam extraction.After simulated in vitro digestion,the original Auricularia auricula polysaccharide was degraded.The triple helix structure in the Auricularia auricula polysaccharide disappeared after digestion by Congo red experiment.FT-IR spectroscopy and uronic acid detection proved that uronic acid in polysaccharide was hydrolyzed.Afterwards,the digestion products extracted in vitro were processed on RAW264.7 cells,and it was found that the Auricularia auricula polysaccharide after digestion with gastrointestinal juice can stimulate the proliferation of macrophages by stimulating the TLR4 receptor-mediated JNK,ERK and NF-κB signaling pathways,and therefore inducing the release of NO and increasing the phagocytic activity of macrophages.In conclusion,we have reason to believe that Auricularia auricula polysaccharides have good prospects as immunomodulators.
