摘要:利用脂肪酶在非水相体系中催化的对映选择性酯化反应,对外消旋手性药物中间体(±)-1-氯-3-(1-萘氧)-2-丙醇(简称萘氧氯丙醇)进行了拆分,考察了溶剂、酰化剂和底物浓度对酶活性和选择性的影响。结果表明,当溶剂为异辛烷,酰化剂为醋酸酐、底物浓度为氯醇100mmol/L、酸酐50mmol/L时,酶显示出最佳的催化活性和选择性,产物酯的光学纯度可达90%。
Efficient kinetic resolution of a racemic chlorohydrin, (±) 1 chloro 3 (1 naphthyloxy) 2 propanol, was achieved by lipase catalyzed enantioselective esterification in non aqueous media. The best solvent is isooctane and the best acylating agent is acetic anhydride with respect to either the catalytic activity or the enantioselectivity of the enzyme. The optimum concentrations of substrates were shown to be chlorohydrim 100 mmol/L and anhydride 50 mmol/L respecively.
