摘要:Zymomonas mobilis 31821(pZB5) is a plasmid-bearing recombinant strain that can produce ethanol from both xylose and glucose. The stability of plasmid-bearing for the strain is significantly important to its utilization in industry. In this study, the fraction of plasmid-bearing for the strain at different conditions and the maximum specific growth rates for the plasmid-bearing and plasmid-free cell in both glucose and xylose mediums were obtained. The simulation experiment for scaling up from 1 mL to 80 t showed that the fraction for plasmid-bearing cell is 81.0% after 26.3 generations. In addition, it is approximately 90.3% when the strain multiplied in a medium containing 20% wood-dust hydrolysate solution by HCl from a colony to 10 mL. The maximum specific growth rates in glucose medium for the plasmid-bearing and plasmid-free cell are mu(max)(+) = 0-185 h(-1) and mu(max) = 0.195 h(-1) respectively. The probability to produce plasmid-free cell for one division is 0.729%.
