摘要:本文研究了产青霉素酰化酶的大肠杆菌ATCC11105在PE620000-DextranT70两水相系统中的动力学行为,并与在磷酸盐缓冲液中的动力学行为进行了对照。产青霉素酰化酶的菌体在两水相体系中的K_m值为4.35mmol/L,最适pH值为8.0,最适温度为55℃,最稳定pH值为6.0~7.0,最稳定温度为30℃;在磷酸盐缓冲液中K_m值为7.20mmol/L,最适pH值为8.0,最适温度为65℃,最稳定pH值为8.0,最稳定温度为35℃。
The kenitic behavior of E. coli ATCC 11105,a producer of Penicillin acylase,in PEG 20000-Dextran T70 aqueoiis two-phase syst-em has been studied With 0.lmol/L phosphate buffer as control system.Km in the aqueous two-phase system is 4.35mol/L,the optimum pH is 8.0,the optimum temperature is 55℃,the most stable pH is 8.0 and the most stable temoerature is 306℃.While in the control system,they are7.20 mmol/,8.0,65℃,6.0~7.0,35℃ respectively.These data are us-eful to be used in the investigation of the bioconversion of Penicillin to 6-ApA in the aquous two-phase system.
