摘要:目的:以对乙酰氨基酚为模型药,卡波姆(Carbopol)聚合物为骨架材料制备骨架缓释片,对其体外释放度及其药动学进行考察。方法:以HPLC法为分析方法,采用相似因子法评价Carbopol骨架缓释片体外释放行为,并考察其家兔药动学。结果:Carbopol骨架缓释片的释药机制表现为非Fick扩散,释药机制是Fickian释药机制以及从Fickian释药机制向零级释药机制的转化。Carbopol聚合物是影响药物释放的主要因素,释放介质的pH值和片剂硬度对上述缓释片的释放有一定的影响。结论:Carbopol聚合物是优良的高分子材料口服片剂药用辅料,具有较好的开发、应用前景。
OBJECTIVE To evaluate the factors affecting the sustained release in vitro of Carbopol matrix sustained release tablets,Carbopol sustained release tablets were designed and prepared by using paracetamol as model drug.The evaluations in vitro and in vivo were carried out as well.METHODS HPLC method was used as analytical approach.The similarity factor was used to evaluate release profile of Carbopol matrix sustained release tablets in vitro.The studies of pharmacokinetics of Carbopol matrix sustained release tablets in rabbits were investigated.RESULTS The drug release mechanism followed the conversion from Higuchi equation to zero-order equation.The pH environment of the dissolution media was the most important factors affecting the drug release profile of Carbopol matrix tablets.The quantity of Carbopol polymers and the hardness had some influence on drug release profile.CONCLUSION Carbopol polymers can be successfully formulated into a variety of different commercial matrix tablet forms.
