摘要:考察了分离方法、血浆(血清)、渗透压、细胞因子的保存等因素对脐血造血细胞体外培养和检测的影响。结果表明,用明胶沉降加密度梯度离心分离两步法分离比传统方法能分离到更多的单个核细胞;混合脐血血浆比胎牛血清更能支持造血细胞的生长;高渗透压对集落形成有一定影响,而低渗透压对集落形成更为有害;在37°C下保存细胞因子,其生物活性下降最快,-30°C冷冻保藏较好地保持其活性,解冻次数对生物活性影响不显著。
Cord blood (CB) is an alternative source of hematopoietic stem cells. Ex vivo expansion of the CB stem/progenitor cells has a significant potential for clinical applications in transplantation. The effects of some operational factors, such as mononuclear cell separation methods, CB plasma, inoculum density, osmotic pressures , and the preservation of cytokines, on the CB stem/progenitor cell cultures ex vivo were investigated. It was found that gelatin sedimentation plus Ficoll centrifugation separation could reduce the loss of mononuclear cells significantly, and was demonstrated to be more practical, effective and economical than conventional method. Mixed CB plasma was shown to be much more favorable than fetal calf serum and autologous CB plasma in supporting colony forming units (CFU). Hyper osmotic pressure influenced the CFU's output, while the experimental results indicated that hypo osmotic pressure was much more harmful. In contrast to 37°C, under cryopreserved conditions(-30°C) the activities of cytokines could be retained well, and their activities appeared to be independent of the times of defreezation.
