摘要:采用摇瓶和封闭式光生物反应器进行了蓝藻基因工程常用宿主系统集胞藻Syne chocystis680 3的混合营养培养研究。在摇瓶中对集胞藻 680 3利用有机碳源特性进行研究的结果表明 ,光照是集胞藻 680 3利用葡萄糖的必需条件 ,混合营养培养是集胞藻 680 3的最佳培养方式。以葡萄糖为基质 ,在 2 5L封闭式光生物反应器中经过 58.5h的混合营养培养 ,集胞藻 680 3的藻细胞密度和平均生长速率分别达到 2 .50g/L和 1 .0 1 g/L/d,分别是光自养培养同期的 9倍和 1 1倍 ,这表明封闭式光生物反应器混合营养培养方式在促进集胞藻 680 3生长上有显著作用。
Studies on the mixortophic cultivation of Synechocystis sp. PCC6803 in shake flasks and a 2.5L enclosed photobioreactor were carried out. The results from mixotrophic cultivations in shake flasks showed that light was necessary for mixotrophic growth of Synechocystis sp. PCC6803; compared with sucrose, glucose was a better kind of substance for Synechocystis sp. PCC6803 mixotrophically growing. In 2.5L enclosed photobioreactor, after 58.5h mixotrophic cultivation, 2.50g/L cell density and 1.01g/L/d were achieved, which were respectively 9 times and 11 times as much as those in photoautotrophic cultivation. On the basis of above results, it was concluded that mixotrophic cultivation system had an advantage over photoautotrophic cultivation system.
