摘要:NOVELTY - A gene knockout recombinant vector, including Gluconobacter oxydans gene homologous recombinant front arm, gene marker and mitochondrial FAD-linked glycerophosphate dehydrogenase (mgdh) gene homologous recombination back arm, is new. USE - The gene knockout recombinant vector is useful for preparing the gene knockout bacterium of G. oxydans. The gene knockout bacterium of G. oxydans is used as biocatalyst in biosynthesis and chiral synthesis fields (all claimed). ADVANTAGE - The gene knockout bacterium of G. oxydans is used as biocatalyst in biosynthesis and chiral synthesis fields at low cost. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are: (1) a gene knockout bacterium of G. oxydans, where mgdh is gene knockout bacterium; and (2) a method for preparing the gene knockout bacterium of G. oxydans by (a) transforming the gene knockout recombinant vector above into Escherichia coli to obtain the transformant; and (b) jointing two parents of transformant as donor bacterium and the G. oxydans as recipient bacterium, where the recombinant vector enters in the G. oxydans so that the mgdh gene on chromosome is homologously and doubly exchanged to obtain zygote; the zygote is mgdh gene knockout bacterium.
