摘要:采用超滤-纳滤联用技术对出芽短梗霉发酵产生的聚苹果酸进行分离纯化。优化的工艺条件为:选用截留分子量为10 ku的超滤膜进行超滤,温度45℃,操作压力0.4 MPa,洗脱4次,此时聚苹果酸透过率为78.1%;采用截留分子量为300 u的纳滤膜进行纳滤,纳滤温度50℃,操作压力0.8 MPa,洗脱4次,此时离子去除率为81.6%。在超滤和纳滤的最优条件下对发酵液中的聚苹果酸进行分离纯化,最终聚苹果酸得率为67.6%,纯度为92.3%。
To optimize the isolation and purification process of poly malic acid from Aureobasidium pullulan fermentation broth by ultrafiltration-nanofiltration coupling technology. The poly malic acid of 78.1% was obtained under the following optimal condition: ultrafiltration membrane of 10 ku, temperature of 45 ℃, ultrafitration pressure of 0.4 MPa and elution times with water of 4. The removing rate of ions reached 81.6% under nanofiltration membrane of 300 u nanofiltration, temperature of 50 ℃, pressure of 0.8 MPa and elution times with water of 4. To separate and purify poly malic acid from fermentation broth under the optimal conditions of ultrafiltion and nanpfiltration. Finally, the poly malic acid yield was 67.6%, the purity of poly malic acid was 92.3%.
