摘要:采用酸法从牛皮中提取胶原蛋白,研究酸提介质、物料比、提取温度和时间对胶原蛋白提取的影响。实验结果确定的最佳酸法提取牛皮胶原蛋白条件为采用0.5mol/L乙酸作介质,料液比1∶30,32℃下提取6h。经等电点沉淀并纯化后的胶原蛋白结合SDS-PAGE电泳和DSC分析,并与SigmaⅠ型胶原蛋白比较,确定所提取的胶原蛋白为典型的Ⅰ型胶原蛋白。此外,圆二色谱测试和体外成纤维试验结果,进一步表明所提取的胶原蛋白保持了天然胶原的三螺旋结构。
Collagens were extracted from bovine skin by acid method, the effects of the ration of material to liquor, temperature and extract time were studied. The optimum extraction conditions were obtained as follows: 0.5mol/ L acetic acid as extraction media, the ration of material to liquor 1 : 30, and extraction at 32℃ for 6 h. Collagens by isoelectric precipitation and purified were then analyzed by SDS-PAGE and DSC. Comparing with type Ⅰ collagen from Sigma, the extractives were identified as the typical type Ⅰ collagen. In addition, the CD and fibril formation experiment confirmed that the extracted collagens retain its original triple helical structure.
