摘要:NOVELTY - Method for preparing cefprozil involves preparing polymer PADBA, poly-acetoacetyl-L-diaminobutyric acid (PADB), and poly(10-methacryloyloxydecyl phosphorylcholine-co- butyl methacrylate (BMA)) (PMDB) solution in aqueous two-phase system, mixing with e.g. 7-amino-3-propenylcephem-4-carboxylic acid(7-APRA), adjusting pH, adding e.g. immobilized penicillin acylase, reacting, adjusting pH of the reaction liquid, recovering aqueous two-phase system PADBA-PMDB, and PADB-PMDB polymer, crystallizing the mixture, centrifuging, and washing and drying the crystal. USE - The method is useful for preparing cefprozil (claimed). ADVANTAGE - The method is cost-effective, simple and easy, and enables to obtain cefprozil with high yield, and without using any chemicals. DETAILED DESCRIPTION - Method for preparing cefprozil involves (a) preparing polymer PADBA, poly-acetoacetyl-L-diaminobutyric acid (PADB), and poly(10-methacryloyloxydecyl phosphorylcholine-co- butyl methacrylate (BMA)) (PMDB) solution, and uniformly mixing the solution in aqueous two-phase system, (b) uniformly mixing 7-amino-3-propenylcephem-4-carboxylic acid(7-APRA), and D-p-hydroxyphenylglycine methyl ester hydrochloride in a molar ratio of 1:1-1:4, where the solid-liquid is 1:10-1:40, adjusting the pH of the solution to 5-6.50, and maintaining temperature of the solution at 10-30 degrees C, (c) adding immobilized penicillin acylase to the solution, uniformly stirring and reacting the mixture for 2-20 hours, (d) allowing the solution to stand under static condition, removing the immobilized penicillin acylase, adjusting pH of the reaction liquid, recovering aqueous two-phase system PADBA-PMDB, and PADB-PMDB polymer, centrifuging to obtain supernatant, adding N,N-dimethylformamide to the supernatant in a volume ratio is 1:3-1:10, crystallizing the mixture at 0-10 degrees C for 0.5-2.5 hours, adjusting the pH of the crystal to 5-8, then centrifuging the crystal, and washing and drying the crystal.
