摘要:以螺旋链霉菌25-1为出发菌株,分别采用紫外线(UV)、Co60射线、甲基磺酸乙酯(EMS)、氯化锂(LiCl)、UV+LiCl、亚硝基胍(NTG)+UV进行诱变筛选,比较各方法的致死率、正变率、最大提高幅度及对传代稳定性的影响,结果发现UV、Co60射线、UV+LiCl 3种方法对螺旋链霉菌的诱变效果较好,化学诱变剂对螺旋链霉菌的稳定性影响较大;同时采用丙二酸钠和乙酰胺进行高通量筛选,丙二酸钠不但筛选效果好,且对效价有一定的促进作用,乙酰胺对菌株稳定性有一定的影响。
With Streptomyces spiramyceticus 25-1 as primitive strain,ultraviolet(UV),Co60-ray,ethyl methane sulphonate(EMS),lithium chloride(LiCl),UV+LiCl,nitrosoguanidine(NTG)+UV were used for mutation screening.By comparing the death rate,positive rate,maximum improve percentage and the stability,UV,Co60-ray and UV+LiCl exhibited better mutation effect on Streptomyces spiramyceticus,and chemical mutagenesis was not good for the stability.In addition,malonate sodium and acetamide were used for high-throughput screening.The result showed that,malonate sodium could not only screen the strains well,but also promote the titer to a certain degree,while acetamide exerted some influence on the stability of the strains.
