摘要：NOVELTY - A (S)-alpha-(2'-chlorphenyl)-6, 7-dibenzothiophene (3, 2-c) pyridine-5(4H)-acetic acid preparation method involves esterifying amino-(2-chloro-phenyl)-acetic acid (I) to obtain a 2-chloro-benzylamine substituted ester compound (II), which is divided to obtain 2-chloro-benzylamine substituted ester compound (III). The compound (III) is reacted with 2-(2'-thienyl) ethanol tosyl ester and alkali to obtain ((R)-1-(2-Chloro-phenyl)-ethyl)-(2-thiophen-2-yl-ethyl)-amine compound (IV). The compound (IV) is processed under mannich reaction. USE - Method for preparing (S)-alpha-(2'-chlorphenyl)-6, 7-dibenzothiophene (3, 2-c) pyridine-5(4H)-acetic acid for detecting clopidogrel product quality. ADVANTAGE - The method enables preparing (S)-alpha-(2'-chlorphenyl)-6, 7-dibenzothiophene (3, 2-c) pyridine-5(4H)-acetic acid with high purity in a simple, easy and cost effective manner. DETAILED DESCRIPTION - A (S)-alpha-(2'-chlorophenyl)-6, 7-dibenzothiophene (3, 2-c) pyridine-5(4H)-acetic acid preparation method involves esterifying amino-(2-chloro-phenyl)-acetic acid (I) to obtain a 2-chloro-benzylamine substituted ester compound of formula (II), which is resolved to obtain 2-chloro-benzylamine substituted ester compound of formula (III). The compound (III) is reacted with 2-(2'-thienyl) ethanol tosyl ester and alkali to obtain ((R)-1-(2-chloro-phenyl)-ethyl)-(2-thiophen-2-yl-ethyl)-amine compound of formula (IV). The compound (IV) is processed under mannich reaction to obtain 5-((R)-1-(2-Chloro-phenyl)-ethyl)-4,5,6,7-tetrahydro-thieno(3,2-c)pyridine compound of formula (V). The compound (V) is processed under hydrolysis reaction to obtain the (S)-alpha-(2'-chlorphenyl)-6, 7-dibenzothiophene (3, 2-c) pyridine-5(4H)-acetic acid. R=linear or branched at least 2C alkyl, where the configuration of Casterisk is in R shape or S shape.
