摘要:对含有蛹虫草Cu,Zn-超氧化物歧化酶(cm-SOD)基因的重组E.coli培养条件进行了初步优化。结果表明,LB培养基国含有Cu^(2+)可明显提高超氧化物歧化酶比活力;而在菌体培养中补加葡萄糖,可提高重组蛋白表达量和细胞生物量。在含有0.6mmol/L Cu^(2+)、Zn^(2+)的LB培养基中补加22mmol/L葡萄糖,每200ml培养液可获得lg湿细胞,超氧化物歧化酶比活力达到3305u/mg。
The culture conditions for expression of Cordyceps militaris Cu, Zn-superoxide dismutase in E.coli were optimized. The results showed that adding Cu^2+in culture medium could increase the specific activity of superoxide dismutase in E.coli, whereas supplement with glucose during shake flask incubation could increase the amount of biomass and recombinant protein. One gram of wet cell weight could be obtained from 200ml culture when LB medium cantaining 0.6mmol(L Cu^2+ and Zn^2+ and supplementing with 22mmol/L glucose was used, and specific activity of superoxide dismutase was 3305u/mg.
