摘要:用大孔吸附树脂从红霉素发酵滤液中提取、分离红霉素,研究其吸附、洗脱过程,包括吸附剂、洗脱剂的筛选及吸附条件和洗脱条件的选择等。实验结果表明,筛选到HZ816型吸附树脂对红霉素的静态吸附容量为7.22×104u/g抽干吸附树脂(滤液浓度为1290u/ml),吸附等温线属Langmiur型。在pH9.4,流量为3BV/h的条件下对红霉素发酵滤液的动态吸附容量为1.34×105u/g抽干吸附树脂(滤液浓度为4500u/ml),选用乙酸丁酯进行洗脱,当洗脱液流量为0.5BV/h,2BV洗脱液情况下洗脱率为94.18%。红霉素A及红霉素C在树脂上的穿透曲线表明,红霉素A在吸附过程中存在着竞争优势。
The processes of adsorption of erythromycin on macroporous resin from broth filtrate and its desorption are discussed in the present paper. The results showed that the selected HZS16 macroporous resin has static capacity of erythromycin as high as 7.22 × 10^4u/g wet resin from a broth filtrate concentration of 1290u/ml, the adsorption data are agree with the Langmiur isotherm. The dynamic capacity of the adsorption column is 1- 34×10^5u/g (wet resin) from a broth filftrate concentration of 4500u/ml when the operation conditions were as follows: pH9.4, flow rate: 3BV/h, and eluted with butyl acetate as eluant at a flow rate 0. 5 BV/h and elution volume of 2BV, the elution rate was 94.18%. The breakthrough curve of erythromycin A and C reveals that EMA is competitively advantageous over EMC during the adsorption process.
