摘要:NOVELTY - Preparing enzyme-coupled chitosan nanoparticles involves crosslinking chitosan with an enzyme chemically to form enzyme-crosslinked complex, where the isoelectric point of the enzyme is greater than 5. The enzyme refers to horseradish peroxidase, lysozyme, catalase, and lipase. An ionic gel of chitosan nanoparticles is prepared by adding dropwise sodium tripolyphosphate to the obtained enzyme-crosslinked complex on a magnetic stirrer to obtain homogeneous suspension. USE - Method for preparing enzyme-coupled chitosan nanoparticles used for providing enzyme-activating prodrug therapy (claimed). ADVANTAGE - The high biocompatibility and non-toxic enzyme-coupled chitosan nanoparticles are capable of loading good amount of drug and enzyme, and is also prepared in a simple and convenient manner. The (HRP) horseradish peroxidase-encapsulated chitosan nanoparticles for enzyme-prodrug cancer therapy effectively avoids the HRP degradation in the human body and thus controls the release of chitosan nano-particles to certain degree. DETAILED DESCRIPTION - Preparing enzyme-coupled chitosan nanoparticles involves crosslinking chitosan with an enzyme chemically to form enzyme-crosslinked complex, where the isoelectric point of the enzyme is greater than 5. The enzyme refers to horseradish peroxidase, lysozyme, catalase, and lipase. An ionic gel of chitosan nanoparticles is prepared by adding dropwise sodium tripolyphosphate to the obtained enzyme-crosslinked complex on a magnetic stirrer to obtain homogeneous suspension. The obtained homogeneous suspension is washed and then subjected to centrifugation to obtain purified enzyme-coupled chitosan nanoparticles as the finished product.
