摘要:NOVELTY - An isovaleryl spiramycin III is prepared by preparing isovaleryl spiramycin III in spiramycin raw materials using high-speed counter current chromatography; and preparing stationary phase and mobile phase using a solvent system comprising n-hexane, ethyl acetate, methyl alcohol, and water with volume ratio of 1:1-2.3:1.2-2.7:0.8-2. USE - Method for preparing an isovaleryl spiramycin III (claimed). ADVANTAGE - The method is high in speed, fast and free of loss of sample, and suitable for high-speed counter current chromatograph in various forms in preparing isovaleryl spiramycin III. DETAILED DESCRIPTION - Preparation of isovaleryl spiramycin III involves extracting pure spiramycin in spiramycin fermentation liquor by filtering, extracting, reverse extracting, washing, and crystallizing to obtain spiramycin with not less than 25% isovaleryl spiramycin III; preparing n-hexane, ethyl acetate, methyl alcohol, and water into solvent system of stationary phase and mobile phase; pumping the stationary phase in the high-speed counter current chromatograph for filling the whole spiral pipe; starting the host machine to make the spiral pipe rotate high-speedily; pumping mobile phase at 2-3 ml/minute in the high-speed counter current chromatograph; creating balance, pouring the spiramycin by an injection valve; and collecting effluent liquid according to detector spectrogram to obtain isovaleryl spiramycin III. The volume ratio of n-hexane, ethyl acetate, methyl alcohol to water is 1:1-2.3:1.2-2.7:0.8-2.
