摘要:分析酵母基因工程菌生产目的基因表达产物——乙型肝炎表面抗原的发酵过程,降低培养基中葡萄糖浓度,补充甘油和蔗糖,能使乙型肝炎表面抗原相对浓度从3.89提高至8.12,比活值增加2.69倍。根据实验观察的结果,造成发酵过程的温度序列,能合理地分配受体细胞代谢能量的消耗,使目的基因的表达量提高至13.51相对浓度。同时,对流加式操作系统中表现出的重组质粒稳定性的变化规律给予描述和解释。另外,提出了异源DNA与受体细胞的转化过程存在着一个分布,此分布对通过发酵提高乙型肝炎表面抗原的产量造成影响。
Fermentation was performed to produce exogenous protein-hetatitis B surface antigen (HBsAg) by recombinant ycast S. cerevisiae containing the HBsAg gene. These studies showed that relative concentrations of HBsAg were increased from 3.89 to 8.12 when glucese content was reduced and glycerol and sucrose were complemented in culture media, and also, specific activity was increased 2.09-fold. According to the results of experiments, the sequence of temperature variation in fermentation processes was set up. Then the expressed amount of HBsAg gene achieved 13.51 in relative concentration. The stability of a recombinant plasmid containing HBsAg gene was observed and explained by altering fermentation temperatures in fed-batch operation. On the other hand, there is a distribution within transformation between exogcnous DNA and host yeast and it will affect the production of HBsAg by recombinant yeast in fermentation processes.
