摘要:利用Plackett-Burman设计法析出影响必特螺旋霉素效价的四个最显著的因素,以Box-Behnken设计法及响应面分析法确定了主要因素的最佳浓度,即鱼粉22.8g/L,酵母粉2.44g/L,CoCl29.25mg/L,NaCl 9.13g/L,此时必特螺旋霉素效价为2245μg/ml。新配方验证结果表明必特螺旋霉素效价和必特螺旋霉素组分中总异戊酰基螺旋霉素的含量在96h分别达到2380μg/ml和54%,比原配方分别提高了53%和20%。
Four nutritional factors that influenced the titre of carrimycin most significantly were screened out using Plackett-Burman experimental design, and then optimized by Box-Behnken factorial design of response surface methodology in shake flask cultures. Maximum carrimycin titer of 2245 μg/ml was predicted in the medium containing fish meal 22.8 g/L, yeast powder 2.44 g/L, COCl2 9.25 mg/L, NaCl 9.13 g/L using response surface plots and point prediction tool of DESIGN EXPERT7.0 software. Compared to the culture with original medium, the titer and relative content of total isovalerylspiramycins with the optimized medium were increased by 53% and 20% in 96h respectively.
