摘要:NOVELTY - Purifying Grifola frondosa polysaccharide comprises: (i) crushing the Grifola frondosa, extracting the crushed particles via microwave using deionized water to obtain polysaccharide extract; (ii) soaking the resin in deionized water, sodium hydroxide solution, hydrochloric acid solution, and deionized water respectively; (iii) packing the column with treated resin to form a resin bed, allowing it to stand overnight, passing deionized water via column; and (iv) loading the polysaccharide extract into the activated resin column, collecting the eluate and removing protein. USE - The method is useful for purifying Grifola frondosa polysaccharide (claimed). ADVANTAGE - The method is simple, short, economical and environmentally-friendly; has high efficiency with less energy consumption; and suitable for large scale industrial production. DETAILED DESCRIPTION - Purifying Grifola frondosa polysaccharide comprises: (i) crushing the Grifola frondosa, taking 20-meshes crushed particles and extracting via microwave using deionized water at pH 6 to obtain polysaccharide extract; (ii) repeatedly soaking the resin in deionized water, then soaking with sodium hydroxide solution for 18-24 hours, soaking with hydrochloric acid solution for 18-24 hours, and rinsing with deionized water; (iii) packing the column with treated resin to form a resin bed, allowing it to stand overnight, passing deionized water through the column for inactivation before use; and (iv) loading the polysaccharide extract into the activated resin column, collecting the eluate and removing protein.
