摘要:以粘红酵母(Rhodotoru la g lutin is)Q 0402为出发菌株,经紫外诱变,得到一株粘红酵母QX 0402。5 L发酵罐的发酵液中3′-磷酸二酯酶酶活力可达250 U/mL,为文献报道过的最高酶活力的2.5倍。所产生的3′-磷酸二酯酶在70°C下保温1 h,酶活力仍有50%以上。该酶最适反应pH为5.4,最适反应温度为60°C,反应3 h的反应转化率为56.37%,底物浓度可以提高到3%,生产效率为文献报道的3倍。
A Rhodotorula glutinis QX0204 which could produce 250 U/mL thermostable 3′-phosphodiesterase in a 5L fermenter was obtained from Q0204 through ultraviolet irradiation. Being incubated at 70 ℃ for 1 h 3′-phosphodiesterase activity still attained over 50%. Its optimal pH and temperature was 5.4 and 60℃ respectively. The yield of 3′-ribonucleotides reached 56. 37% at 60℃ after 3 h reaction. Substrates concentration used could be increased to 3′ and the productivity was 3 times more than that reported.
