摘要:大肠杆菌 JL3是 JM1 0 1经诱变和连续培养所选育到的乙酸耐受性突变株。 JL3在复合培养基中产生的乙酸比 JM1 0 1少 ,菌体对葡萄糖的得率 ( YX/ S)增加。在基本培养基 M中连续培养 ,最大菌体得率 ( YG)增大 ,维持系数 ( m)降低。存在外源乙酸时 ,JL3对葡萄糖亲和性增加 ,显示较强的生长优势。在含有 1 0 g/L乙酸钠的复合培养基中 ,JL 3/p UC1 9的β-半乳糖苷酶比活比 JM1 0 1 /p UC1 9提高
Escherichia coli JL3, a mutant with improved acetate tolerance, was selected from Escherichia coli JM101 by means of mutation and subsequent continuous cultivation with acetate as a selective pressure. In batch cultures in complex media, acetate formation by JL3 was lower than that by JM101, while its cell yield based on glucose, Y X/S , was increased. Data obtained from continuous cultures in a defined medium suggested an improved maximum cell yield( Y G) and a lower maintenance coefficient of JL3. JL3 showed a higher affinity to glucose and distinct growth advantage in media containing acetate. In a complex medium containing 10 g/L sodium acetate, the specific activity of β galactosidase expressed in JL3/pUC19 was increased by 30% than that in JM101/pUC19.
