摘要:目的研究贯叶金丝桃中苯并二蒽酮类成分的超声提取以及大孔吸附树脂纯化工艺。方法采用单因素实验研究超声提取条件,以每克药材中金丝桃素和伪金丝桃素提取量之和为指标,考察了超声提取中影响苯并二蒽酮类成分提取量的因素。通过静态吸附和动态吸附实验选择分离介质,并对所选树脂的分离工艺进行研究。结果贯叶金丝桃中苯并二蒽酮类成分最优提取条件为:15倍量70%的乙醇超声提取2次,每次40min。选取D101大孔树脂为分离介质,上样流速为0.5 BV/h,以6BV 70%乙醇以2BV/h流速洗脱目标物,最终产品中含苯并二蒽酮类成分约6.3mg/g。结论该贯叶金丝桃中苯并二蒽酮类成分的超声提取和大孔树脂分离富集工艺操作条件温和,稳定可靠。
Objective To establish a method for ultrasonic extraction and separation of naphthodianthrones from Hypericum perfo- ratum by macroporous resins. Methods In the optimization of the conditions of uhrasonic extraction, hypericin and pseudohypericin were selected as targets representing naphthodianthrones in the herb. Factors affecting the amount of naphthodianthrones in the extact were investigated. Qualified macroporous resins were chosen based on their performances in both static and dynamic adsorption. Meanwhile,other conditions affecting the separation were probed as well. Results The optimal conditions for extraction and separation are as following:The targets were twice uhrasonically extracted by 15 -fold of 70% aqueous ethanol for 40 rain each time. Then,the extract was loaded on to a D101 resins column at 0.5 bed volume ( BV)/h. The targets were eluted by 6 BV of 70% aqueous ethanol at 2 BV/h ,which yielded naphthodianthrones as 6.3mg/g in the final extract. Conclusion The developed method is reliable for ultrasonic extraction and separation of naphthodianthrones from Hypericum perforatum by macroporous resins.
