摘要:考察HZ-806,HZ-807,HZ-818,HZ-18,HZM-3树脂对发酵上清液中Nisin Z的吸附和解吸性能,筛选出HZM-3树脂来富集浓缩Nisin Z。工艺条件是以20BV/h流速上样,用体积分数为70%的乙醇溶液以2BV/h流速洗脱Nisin Z。吸附过程为放热过程,且更符合Langmuir方程,前期和后期主要吸附速率控制步骤分别是液膜扩散和颗粒内扩散。一轮吸附解吸后Nisin Z纯度提高了95.18倍,回收率为96.7%。
Five macroporous adsorption resins,including HZ-806,HZ-807,HZ-818,HZ-18,HZM-3 are studied on the enrichment and purification properties of Nisin Z in the fermentation supernatant. HZM-8 is selected to enrich Nisin Z according to the experimental results.The optimum dynamic adsorption and desorption conditions for Nisin Z are as follows:flow rate is 20 BV/h for adsorption and 2 BV/h for desorption;70% of ethanol aqueous solution (V/V)as eluant;the thermodynamic parameters indicate that the adsorption is an endothermic process and it could be well described by Langmuir equation.The adsorption rates of the initial stage and the later stage are controlled by liquid film diffusion and intraparticle diffusion.The purity of Nisin Z after adsorption and desorption is im-proved by 9 5 .1 8 folds and the recovery efficiency is 9 6 .7%.
