摘要:NOVELTY - Recombinant human bone morphogenetic protein-2 (rhBMP-2)-loaded chitosan microspheres comprise chitosan microspheres as substrate and rhBMP-2, which is adsorbed on chitosan microspheres, as loaded medicine. The mass ratio of rhBMP-2 and chitosan is 1-30:100. USE - rhBMP-2-loaded chitosan microspheres used for curing bone injury (claimed), bone defect, and delayed union of bone; and for uniting bones. ADVANTAGE - The preparation of rhBMP-2-loaded chitosan microspheres is simple; and has moderate preparation condition, thus, keeping the bioactivity of rhBMP-2. The rhBMP-2-loaded chitosan microspheres can control the release of rhBMP-2 for a long time, thus, accelerating the formation of new bone. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for preparation of rhBMP-2-loaded chitosan microspheres comprising dispersing chitosan solution to oil phase with surfactant to form water-in-oil (W/O) type emulsion system; adding ion crosslinking agent solution to W/O type emulsion system and reacting for 1-12 hours; adding acetone; collecting chitosan microspheres; dispersing chitosan microspheres into water after washing with acetone and absolute ethyl alcohol; dropping 0.1-5 wt/vol.% sodium hydroxide solution while slowly stirring until the final pH is greater than 7; collecting chitosan microspheres; washing with water or dialyzing in water and freeze drying, or soaking chitosan microspheres in 0.01-1 wt/wt.% wt/sulfuric acid aqueous solution for 1-60 minutes, washing with water or dialyzing in water, and freeze drying; dispersing chitosan microspheres in 0.1-10 mg/ml rhBMP-2 solution; passively adsorbing and loading for 0.5-12 hours; and directly freeze drying the obtained dispersion to obtain rhBMP-2-loaded chitosan microspheres.
