摘要:NOVELTY - Culturing ferment seed liquid by talking solid straw as carbon source comprises activating the ferment strain; adding the pre-processed solid straw in the culture medium to synchronously saccharify, domesticate and culture the seeds; multi-level expanding process of the seed liquid; and inoculating and fermenting. USE - The method is useful for culturing ferment seed liquid by talking solid straw as carbon source (claimed). ADVANTAGE - The method has the following advantages: eliminates the infected strain risk in the process of solid-liquid separation to obtain the hydrolyzing liquid; improves the durability of the strains on a suppression object and a solid substrate; process cost is reduced; fermentation period is shortened; and has good industrial application prospect. DETAILED DESCRIPTION - Culturing ferment seed liquid by talking solid straw as carbon source comprises (a) activating the ferment strain: inoculating the strains saved in a strain keeping flat plate, a keeping inclined face or a freezing glycerin keeping pipe to a synthesis culture medium, where the culturing temperature is 25-45 degrees C; the rotating speed is 50-500 rounds per minute (rpm); the culturing time is 12-36 hours; and the optical density (OD)600 value reaches 4-16; in the synthesis culture medium, the concentration of the glucose is 10-100 g/L; the nutrition salt is potassium dihydrogen phosphate (KH2PO4) or dipotassium phosphate (K2HPO4) with the concentration of 0.5-10 g/l and magnesium sulfate heptahydrate (MgSO4.7H2O) 0.1-5 g/l, ammonium sulfate ((NH4)2SO4) 0.5-10 g/l, and leaven extraction power or corn serum with the concentration of 0.1-10 g/l; the strains comprise Saccharomyces cerevisiae, Pachysolen tannophilus, Pichia stipitis, Kluyveromyces marxianus, and Zymomonas mobilis; the above strains are improved by inducing, gene engineering and the like; and two or more than two kinds of the above strains are mixed and fermented, where the induced and screened strains are the strains improved by chemical, physical and ray induction; the strains of the gene engineering are the strains for metabolizing the hexose or pentaglucose to ferment and produce the ethanol after passing through the paths of transforming the plasmid, reconstructing same source and improving the metabolization; OD600 value is the light absorption value of the liquid of the strains at the wavelength of 600 nm; (b) adding the pre-processed solid straw in the culture medium to synchronously saccharify, domesticate and culture the seeds: inoculating the activated seed liquid with the volume percentage of 5-20% to a mixed culture medium of the solid straw, where the mass percentage of the solid straw is 3-30% in the mixed culture medium; adding the cellulase and nutrition salt in the mixed culture medium with the rotating speed of 50-500 rpm, at 25-45 degrees C to culture for 12-36 hours and obtain the serum of the ferment strain; the mixed culture medium is the culture medium for adjusting the solid content of the solid straw by the 0.1-50 g/L glucose solution, where the pH is 2.0-8.0; the cellulase is selected from the mixed enzyme liquid of one or two of the cellulase or the cellobiose; the enzyme added amount is 1-30 FPU/gDM; FPU is the cellulase filter paper activity in the cellulose liquid; DM is the dry weight of the straw object, where the cellulose liquid comprises the commercialization cellulase and the culture liquid with the cellulase activity cultured by the mycete microorganism; the solution for adjusting pH is selected from the mixed solution of one or two of the sulfuric acid, calcium hydroxide, sodium hydroxide or ammonia water; the nutrition salt is KH2PO4 or K2HPO4 with the concentration of 0.5-10 g/l and MgSO4.7H2O 0.1-5 g/l, (NH4)2SO4 of 0.5-10 g/l, KH2PO4 or K2HPO4 with the concentration of 0.5-10 g/l and leaven extraction power or corn serum with the concentration of 0.1-10 g/l; the solid straw is selected from the solid straw raw material or the solid straw processing object; (c) multi-level expanding process of the seed liquid: inoculating the seed liquid obtained in step (b) to the culture medium containing the solid straw, where the mass percentage of the solid straw in the culture medium is 3-30%; adding the cellulase and the nutrition salt; expanding and culturing the seeds by shaking the bottle or in a seed pot; the culture rotating speed is 100-50 rpm; the temperature is 25-45 degrees C to culture for 12-36 hours; obtaining the seed culture liquid; the culture level in the seed liquid multi-level culture process comprises 1-level to 6-level; the volume percentage of the inoculating amount in each culture level is 5-20%; the seed liquid inoculated to the ferment pot comprises the culture liquid in any level of the obtained from the 1-level to the 6-level for culturing the seeds; obtaining the serum containing the ferment strains; and (d) inoculating and fermenting: inoculating the serum containing the ferment strain obtained in steps (b) and (c) through culturing process to a ferment pot with high solid content to synchronously saccharify, ferment and prepare the ethanol or other organism-based products.
