摘要:金色链霉菌摇瓶培养适宜的接种量为3.2%、接种时间为25h、温度在28°C左右。4g/L的硫酸铵和0.043g/L无机磷有利于产物金霉素(CTC)合成。酵母膏的加入对CTC的合成有一定促进作用,以3g/L的效果较好。在50L发酵罐中,较低浓度的无机磷对CTC发酵有利;酵母膏的加入量以4.2g/L最佳,产物效价比酵母膏加入量为2.8g/L和5.6g/L时高3.1%和9.7%,优化发酵过程的CTC产率提高了18%。
The optimal quantity and age of inoculation and temperature is 3.2%、25 h and around 30 °C respectively in CTC fermentation in flask. The effects of 4 g/L (NH_4)_2SO_4 and 0.043 g/L inorganic phosphate on CTC biosynthesis are better than the others. The supplement of yeast extract benefits to the fermentation and the concentration of 3 g/L is better. The results of CTC fermentation in 50 L fermentor (indicate) the titer of CTC at 0.05 g/L KH_2PO_4 is increased by 6.7% compared with 0.15 g/L. And 4.2 g/L yeast extract is the best to CTC biosynthesis, the CTC titer is higher by 3.1% and 9.7% than other two concentration at 2.8 g/L and 5.6 g/L. The CTC titer in optimized process increased by about 18%.
