摘要:NOVELTY - An optically controlled gene expression system, comprising (a) a photosensitive recombinant transcription factor encoding gene, where the photosensitive recombinant transcription factor comprises first polypeptide as DNA binding domain, second polypeptide as photosensitive domain, and third polypeptide as transcription regulatory domain, and (b) a target transcription unit comprising at least one reaction element identified and bound by the first polypeptide, and a promoter and a nucleotide sequence to be transcribed regulated by the third polypeptide, is new. USE - The optically controlled gene expression system is useful for treating type I diabetes mellitus in mammals (including human) to be treated (claimed). ADVANTAGE - The optically controlled gene expression system has quick, effective and powerful induction, is safer than other inducers, is of little or no toxicity, and can control gene expression both spatially and temporally. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for the following: (1) eukaryotic expression vector comprising the above-cited optically controlled gene expression system; (2) method for regulating expression of gene through applying the optically controlled gene expression system in the host cell, involves (a) constructing the optically controlled gene expression system in a eukaryotic expression vector, (b) introducing the host cell of regulated gene, and (c) irradiating the host cell for induction to express the regulated nucleotide in the host cell; and (3) kit, comprising the eukaryotic expression vector and/or the cells of mammalian which is transfected with the eukaryotic expression vector of transcription factor, and corresponding specification.
