摘要:NOVELTY - A Klebsiella pneumoniae ECU-21 (CGMCC 2850), is new. USE - The K.pneumoniae ECU-21 (CGMCC 2850) is useful for producing hydrogen and 2,3-butanediol (claimed). ADVANTAGE - The hydrogen and 2,3-butanediol are produced by a simple process under anaerobic condition, and the method is applicable to industries. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for method of producing hydrogen and 2,3-butanediol using the K.pneumoniae, involves (a) inoculating K.pneumoniae colony from plane plate to seed culture medium comprising (in g/l): glucose (4.5-5.5), yeast powder (4.5-5.5), peptone (9.5-10.5), beef extract (5.5-6.5), sodium chloride (4.5-5.5), potassium dihydrogen phosphate (0.05) and dipotassium hydrogen phosphate (0.05), sterilizing the culture medium and controlling the pH of the medium at 6-6.2, (b) introducing the culture (30 ml) in shaking bottle (250 ml) and incubating at 37 degrees C for 12 hours, (c) inoculating the seed culture (600 ml) in shaking bottle (1 l) containing fermentation medium comprising glucose (29-31), yeast powder (4.5-5.5), peptone (9.5-10.5), beef extract (5.5-6.5), sodium chloride (4.5-5.5) and phosphate buffer (200 mM, pH 6-6.2), sealing fermentation bottle with rubber plug, inletting nitrogen to form an anaerobic culture environment, and exhausting excess gas from exhaust port, and (d) connecting an air bag to the exhaust port to collect excess hydrogen, and obtaining 2,3-butanediol after exhausting of the gas, where the fermentation is performed at 37 degrees C for 12-15 hours in the presence of stirring (150 rotations/minute) using a magnetic stirrer.
