摘要:目的以戊二醛(GA)为交联剂固定化黏质沙雷菌ECU1010脂肪酶活性包涵体。方法单因素优化固定化条件,以GA浓度、酶浓度、GA与酶用量比、固定化时间4因素3水平正交试验确定最佳固定化条件,并研究固定化酶的操作稳定性和贮存稳定性。结果最佳固定化条件为:GA浓度0.15%(w/v),酶浓度15 mg/mL,GA与酶用量比6:1,固定化时间2 h。此条件下制备的固定化酶平均酶活达234.7 U/L,酶活回收率40%以上。固定化酶重复使用6次后仍能保持50%的酶活。4℃贮存3个月后其酶活基本无明显减弱。结论脂肪酶活性包涵体经固定化后其操作稳定性和贮存稳定性显著提高。
Objective To immobilize lipase active inclusion bodies from Serratia marcescens ECU1010 by cross-linking with glutaraldehyde.Methods The immobilization conditions were optimized by single factor analysis.The optimum immobilization conditions were determined by an orthogonal test with four factors and three levels.Finally,the operation and storage stability of immobilized enzyme were investigated.Results The optimum immobilization conditions were as follows: The concentrations of the glutaraldehyde and the enzyme were 0.15%(w/v) and 15 mg/ mL,respectively.The ratio of the glutaraldehyde and the enzyme was 6:1 and the immobilization time was 2 h.Under the optimum conditions the average activity of immobilized lipase reached 234.7 U/L and the residual activity was more than 40%.After 6 times of repeated use,the activity of the immobilized lipase still kept 50% of its original activity.Almost no activity was lost after being stored at 4℃ for 3 months.Conclusion The immobilized lipase active inclusion bodies showed good operation and storage stability.
