摘要:NOVELTY - A recombinant Alcaligenes sp. strain ECU0401 (CGMCC 2009), is new. USE - The recombinant Alcaligenes sp. strain ECU0401 is useful for producing nitrilase that catalyzes hydrolysis of mandelonitrile to R-(-)mandelic acid. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for the following: (1) nitrilase-producing gene (G1) comprising (a) a 1071 base pair sequence (SEQ ID NO: not defined) fully defined in the specification, or (b) a sequence having the above sequence (a), in which one or more base pairs have been added, deleted or substituted; (2) recombinant plasmid comprising the above-cited gene (G1); (3) preparation of the above-cited recombinant Alcaligenes sp.; (4) recombinant nitrilase comprising (c) a 357 amino acid sequence (SEQ ID NO: not defined) fully defined in the specification or (d) a sequence having the above sequence (c), in which one or more amino acids have been added, deleted or substituted; and (5) method for catalyzing the hydrolysis of mandelonitrile to R-(-)mandelic acid, involves proliferating the recombinant Alcaligenes sp., suspending the proliferated recombinant Alcaligenes sp. in buffer solution or immobilizing the recombinant Alcaligenes sp., then reacting substrate, preferably mandelonitrile or its analog with suspension of Alcaligenes sp. or immobilized Alcaligenes sp., for 1-10 hours at 20-60 degrees C, and finally extracting target from the reaction product.
