摘要:用不同有机溶剂依次对荷叶离褶伞(Lyophyllum decastes)子实体醇提物进行萃取,得石油醚、氯仿、乙酸乙酯、正丁醇萃取物和剩余物。化学定性鉴别结果表明:石油醚萃取物中含有甾类(或三萜)、有机酸类、黄酮类物质;氯仿萃取物中含有生物碱类、甾类(或三萜)、有机酸类、黄酮类、蒽醌类物质;乙酸乙酯萃取物中含有生物碱类、甾类(或三萜)、有机酸类、黄酮类物质;正丁醇萃取物和剩余物中均含有糖类、氨基酸(蛋白)、黄酮类和有机酸类物质。体外抗肿瘤活性研究结果表明:石油醚、氯仿和乙酸乙酯萃取物对K562、HepG2和SW620肿瘤细胞的增殖均有抑制作用,氯仿萃取物对人正常前列腺细胞WPMY-1的增殖也有抑制作用,但在实验浓度内石油醚和乙酸乙酯萃取物对该正常细胞的增殖无抑制作用,实验浓度内的正丁醇萃取物和剩余物仅对肿瘤细胞K562的增殖有抑制作用,对供试的其它肿瘤细胞和正常细胞均无抑制能力。
Lyophyllum decastes fruit bodies(50g)were suspended in 20 vols of 95%ethanol(w/v),extracted for three consecutive 2-hour periods using ultrasound,and the combined extracts were concentrated to200 mL.After further sequential extraction three times with equal volumes of petroleum ether,chloroform,ethyl acetate and n-butanol,the four resultant fractions and the residual ethanolic extract were evaporated to dryness and subjected to qualitative chemical analysis.This indicated the presence of(i)steroids(or triterpenoids),organic acids and flavonoids in the petroleum ether extracts,(ii)alkaloids,steroids(or triterpenoids),organic acids,flavonoids and anthraquinone in chloroform extracts,(iii)alkaloids,steroids(or triterpenoids),organic acids and flavonoids in ethyl acetate extracts,and(iv)saccharides,amino acids(proteins),flavonoids and organic acids in both the n-butanol and the residual ethanolic extracts.Proliferation of K562,HepG2 and SW620cancer cells were inhibited by petroleum ether,chloroform and ethyl acetate extracts,while the n-butanol and residual ethanolic extracts inhibited K562 cell growth.However,chloroform extracts also exhibited cytotoxicity to the human prostate stromal cell line,WPMY-1.
