摘要:目的探讨川陈皮素对Caco-2、HeLa、HepG2、A375、SW1990 5种肿瘤细胞的增殖抑制作用及对HeLa细胞的作用机制。方法 0~200μmol/L浓度的川陈皮素作用于体外培养的肿瘤细胞,应用MTT法检测细胞存活率;流式细胞仪及Hochest33258染色检测细胞凋亡率及形态学变化;分光光度法检测凋亡蛋白酶caspase-3的变化。结果川陈皮素可显著抑制5种肿瘤细胞的生长,且对HeLa细胞的抑制效果最好。川陈皮素可促进HeLa细胞的凋亡,并可以增加其凋亡蛋白酶caspase-3的活性。结论川陈皮素能抑制5种肿瘤细胞尤其是HeLa细胞的生长。川陈皮素可诱导HeLa细胞发生凋亡;提高凋亡蛋白酶caspase-3的活性可能是其诱导细胞凋亡的作用机制之一。
AIM To investigate the anti-tumor effects of nobiletin on five human cancer cell lines Caco-2,HeLa,HepG2,A375,SW1990 and its anti-tumor mechanisms on HeLa cells.METHODS Five established human cancer cell lines in culture medium were treated with 0-200 μmoL/L nobiletin.The cell viability rate was measured by MTT assay.The cell apoptotic rate of HeLa cell was detected by flow cytometry and apoptotic morphology changes were studied by Hochest 33258 staining.The activity of caspase-3 was determined by spectrophotometrical analysis.RESULTS Nobiletin could inhibit the growth of five human cancer cell lines and has best inhibiting effect on HeLa cells.Nobiletin could increase the apoptotic rate of HeLa cells and also could increase the caspase-3 activity.CONCLUSION Nobiletin has apparent antitumor effects on these five human cancer cell lines,especially on HeLa cells,in vitro.Increasing the caspase-3 activity and promoting the apoptosis rate of HeLa cells may be one of its anti-tumor mechanisms.
